An intracellular calcium overload is associated with the overproduction of reactive oxygen species and the subsequent neuronal cell death [[@B35]]. In this study, the intensity of PTZ-induced convulsions was decreased by insulin treatment in a dose-dependent manner. The dose of insulin at 5 nmol/head increased the threshold dose for the development of convulsions, but the inhibition of PTZ-induced convulsions by insulin at 15 and 50 nmol/head was not significant. In addition, insulin treatment decreased the intracellular calcium level in the hippocampus. It has been reported that the insulin-mediated reduction in the calcium level of cerebellar granule cells of rats was antagonized by the PKC inhibitor calphostin C [[@B18]]. Taken together, these findings suggest that insulin attenuated PTZ-induced convulsions by reducing the calcium level in the hippocampus.
