Category:Finnish photographers Category:1961 births Category:Living peopleThe broad, long-term objectives of this proposal are to elucidate the biochemistry and regulation of intestinal cytochromes P450 (CYP450) and the role of CYP450s in modulating intestinal toxicity caused by chemicals and drugs. Cytochromes P450 metabolize a diverse group of drugs, contaminants, and toxins. The majority of intestinal CYP450s are localized to the basolateral membranes of enterocytes in both the small and large intestines. CYP450 activity can increase up to 10 fold in rats when they are subjected to a single dose of 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD). The induction of CYP450 can protect the intestine from damage caused by lipophilic xenobiotics. The inducibility of CYP450s may be important for the intestinal toxicity of drugs and chemicals that are ingested or absorbed. A lack of this induction or a loss of this induction by a variety of factors can lead to drug-induced enteropathy, which is a major source of morbidity and mortality in the United States. The specific aims of this proposal are: 1) Characterize CYP450 induction in the small intestine of rats treated with 2,3,7,8-TCDD and elucidate the mechanisms for CYP450 induction in this tissue. 2) Determine the differences in the expression of CYP450s in the small intestine and liver. 3) Elucidate the role of CYP450s in the intestinal toxicity of chemical xenobiotics in rats. The methods to achieve these aims are kinetic analyses of CYP450 induction, microsomal CYP450 content measurements, and measurement of CYP450 enzyme activities. The kinetics of CYP450 induction will be determined by measuring the expression of mRNAs for specific CYP450 isoforms. CYP450 induction will be characterized by measuring CYP450 content and the activities of relevant CYP450 enzymes. The role of CYP450 induction in intestinal toxicity will be examined by examining the role of CYP450 induction in the intestinal toxicity of selected model chemicals. CYP450 induction in the small intestine will be examined by measuring CYP450 induction in the duodenum, jejunum, ileum, and colon. Furthermore, the induction of CYP450s in the liver and small intestine will be determined. The
