As such, despite effective treatment for HIV infection, many people with HIV have a significant risk of CVD, with approximately 50% dying from CVD. A major factor driving the increase in CVD events in HIV-infected individuals is that HIV infection has been linked to an accelerated atherosclerotic process. Thus, CVD has emerged as a major cause of morbidity and mortality in HIV-infected individuals. The goal of this proposal is to define novel atherogenic mechanisms and pathways that are activated in a mouse model of HIV infection. We and others have defined in detail how HIV replication, chemokines, and proinflammatory cytokines promote lipid infiltration into the arterial wall and activate endothelial cells and smooth muscle cells, and thereby promote atherosclerosis. However, the mechanisms that HIV and chemokines stimulate to activate endothelial cells are largely unknown. We have shown that the CD4+ Th1 lymphocyte cytokine IFN? induces ICAM1 expression on endothelial cells, and that this ICAM1-mediated adhesion is required for endothelial leukocyte transmigration into the arterial wall. Furthermore, we have shown that IFN? stimulates the activation of endothelial cells through the STAT1 pathway. As such, we hypothesize that CD4+ Th
